Abstract :
In this research, the influence of different mutational treatments (Ethidium bromide (EtBr) and acrylamide treatment) on amylase production was studied. EtBr mutant showed increased in alpha amylase activity at 200µg/ml with enzyme activity compared with the wild strain at different incubation time. Increase in EtBr concentration from 200 µg/ml, 300 µg/ml and 400 µg/ml shows increase in enzyme activity 0.603455 U/ml, 0.652753 U/ml and 0.711182 U/ml respectively. Decreased in enzyme activity was observed in EtBr mutant exposed to 500 µg/ml and 600 µg/ml with enzyme activity 0.661883 U/ml and 0.579718 U/ml respectively. Acrylamide mutant showed no increase in enzyme activity at concentration of 200 µg/ml compared with wild strain at different incubation time. Increase in acrylamide concentration from 200 µg/ml, 300 µg/ml and 400 µg/ml shows increase in enzyme activity (0.267492 U/ml, 0.740396 U/ml and 0.818909 U/ml respectively). At concentrations of 500 and 600 the enzyme activity decreased with enzyme activity 0.683793 U/ml and 0.42817 U/ml.
